Forgive me, but aren't those guys already here? I own five different Joe Bonamassa albums. They're awesome, but it's not really up and coming, is it?
I would imagine this category is someone half of us will scratch our heads at the name. I can't think of anyone right now, but I'm sure the forumites will.

I think the next wave of acoustic players like Andy McKee, Tommy Emmanuel, and so on and so forth are really poised to do something in the mainstream (non-guitar obsessed) world.

Brendan Small. His work on The Dethalbum (as Dethklok's guitarist Skwisgaar Skwigelf) shows him not only a very funny man, but also an incidinery metal axeman.

It's not easy to identify "up and coming" guitarists, since the main music industry isn't featuring guitarists much these days.
Derek is indeed exceptional in that he has amazing skills, and the venue to exploit them.
Sadly many great young guitarist are not given the exposure that would make wide knowledge of their skills commonplace.

i don't think trucks is "up and coming.
Arjen Lucassen (Ayreon) is my choice. He is a very talented musician who is becoming more well known is the progressive rock world. he constructs these really epic rock symphonies that i love.

I reviewed the record from Jet Lag Gemini a few months back and their guitarist, Vlad Gheorghiu, is a great player. The band does sort of a pop/punk thing but this guy shreds over the top of it. Very refreshing to hear a young guitarist in that style really know his stuff. Definitely worth a listen.

Geoff Achison -- at least here in the States. It's a crime that he's not more famous than he is. But hopefully his grass-roots tour of the south will change that.

Well, depending on what style you are talking about I know of one guitarist (been around for 6yrs or so) in the country genre who is amazing in his fresh approach to country leads. "Brad Paisley" incorporates jazz elements into his super fast hybrid picking "chickin' pickin'" licks. Totally amazing and fresh from anyone who is out there in that genre today.

I would add Luther Dickinson from North Mississippi Allstars and now the Black Crowes. As with other suggestions here, Luther is far from being "new," but joining the Crowes has certainly upped his visibility and put him in a wider spotlight. His work on the new Crowes disc is stellar, to say the least.
And yeah, Derek Trucks. Beyond words. Perhaps the most otherworldly musician I've ever seen.
